  • Aspergillus niger (ATCC 16404) 

  • Escherichia coli (ATCC 4352) 

  • Pseudomonas aeruginosa (ATCC 9027) 

  • Staphylococcus aureus (ATCC 6538) 

These microbial strains are selected based on their ability to contaminate cosmetic products and their potential to cause infections if not controlled effectively.

Testing methodology

  • Initial count of microorganisms in the test product is determined using the plate count method.  

  • The test product is inoculated with a known concentration of specific test organisms.

  • Depending on the test organism, the inoculated product is incubated under specific conditions for 28 days.

  • During incubation, the number of microorganisms is counted at specific intervals like 7, 14, and 28 days.

  • The test product’s preservative efficacy is determined by comparing the microbial count at specific time intervals to the initial count of test microorganisms.

  • In the case of bacterial testing, post-incubation, the log reduction in the microbial count is determined to confirm the effectiveness of the preservative in reducing or inhibiting microbial growth.

Acceptance criteria

To pass the USP 51 test for cosmetic safety, the preservative system must meet the following criteria:

1. For bacterial strains

  • The number of viable bacterial cells must be reduced by at least 99.9% (2 log reduction) within 14 days.

2. For fungal strains (yeasts and molds)

  • The growth of yeasts and molds should remain at or below the initial contamination level (no increase)​.
