Introduction
The Japanese Industrial Standards (JIS) L 1902 method has been developed to test the antibacterial activity and efficacy of fabrics treated with antimicrobial agents.
Test Components
Designed to test the antimicrobial activity of various fabrics, the JIS L 1902 method includes both qualitative and quantitative components.
The qualitative component of the JIS L 1902 testing method is a qualitative determination of growth on a textile. The quantitative method necessitates an extra incubation period for the determination of the bacterial population by plating.
Methods of evaluation
JIS L 1902 is similar to standard antimicrobial textile test methods that examine textile materials following a brief incubation period. Ideally, this method is used for high throughput, accelerated turnaround, and substantial volume testing.
1. Absorption Method
In this method, the test bacteria is inoculated directly onto the sample. The standard contact time for the JIS L 1902 test method is 18 to 24 hours. The number of bacteria present directly after inoculation and after the stipulated contact time is measured and used to estimate the antibacterial activity.
2. Transfer Method
This evaluation method consists of placing the test bacteria on an agar plate and subsequently transferring them onto the samples. The sample textile materials are kept in contact with the bacteria for five minutes and incubated for 18 to 24 hours. The number of bacteria present directly after inoculation and after the stipulated contact time is measured and used to estimate the antibacterial activity.

3. Printing Method
In this evaluation method, the test bacteria are placed on a filter and printed onto the samples.
4. Halo Method
The textile sample is kept in contact with the surface of these inoculated agar plates. The plates are incubated for 24 to 48 hours and then assessed for “halos”. This is a qualitative method that estimates antibacterial activity through the appearance of halos [clear area of growth] around the sample.
The incubation stipulations of the JIS L 1902 test make it one of the most rigorous methods for evaluating antimicrobial activity. This presents a robust replicate analysis and durability of the treated test sample.

The test protocol
The microbes to be tested are prepared by culturing them in a liquid culture medium. The test organisms are standardized through dilution in a nutritive broth. This broth offers optimal conditions for the microbes to grow. Optimal growth conditions are essential for the test to be conclusive.
The control and test fabric samples are inoculated with the microbes in triplicate. These triplicates are incubated in closed containers at body temperature for 18 hours. The initial number of microbes is estimated at “time zero” through elution, dilution, and plating of the control textiles directly after inoculation.
Post incubation, the final microbial concentrations are measured. The decrease in microorganisms relative to initial concentrations and the control fabric is estimated. The controls used are examined to guarantee that the elution method neutralizes the antimicrobial agent in the test samples.
