EPA MLB SOP MB-41: Standard Method for the Evaluation of Antimicrobial Activity of Hard, Non-porous Copper-containing Surface Products
EPA MLB SOP MB-41 Test determines the efficacy of hard, non-porous Copper-containing surface products with antimicrobial activity against test bacteria. The method is used to assess the antimicrobial activity upon mechanical abrasion of surface coatings containing Copper, on hard, non-porous surfaces against prescribed microbes such as Staphylococcus aureus, Pseudomonas aeruginosa, and Viruses. However, the test can be adapted to other bacterial strains as well.
EPA MLB SOP MB-41 Test Method is divided into 2 parts
Part 1: Abrasion and chemical treatment
- Copper carriers and stainless steel control carriers of size 2.5 sq. cm. are brush-coated with antimicrobial chemicals and act as test specimens, with uncoated carriers acting as controls.
- The carriers are exposed to at least 3 different chemical treatments for any antimicrobial sample, each followed by an abrasion cycle.
- The carriers are exposed to the chemical treatments and abrasion 5 times a day, for 5 days a week, for 6 weeks continuously.
Part 2: Product efficacy
- After the completion of the treatment and abrasion, the carriers are challenged with a 20μL mixture of the test organism and soil load for a contact time of 1-2 hours.
- After the contact time, the antimicrobial action of the test specimen is neutralised using a neutralising media, and the surviving test organism is recovered.
- The number of viable microorganisms recovered from the controls (uncoated and unabraded) and test specimens (coated and aberrated) is determined quantitatively.
- The number of viable microorganisms on treated and/or abraded carriers is compared to the untreated control carriers and the antimicrobial efficacy is determined by log reduction.
A minimum of 3 log reduction in microorganisms within 1-2 hrs indicates a residual claim for the antimicrobial surface coating.

EPA MLB SOP MB-41 testing provides supporting efficacy data for the registration of hard, non-porous Copper-containing surface products used to supplement standard disinfectants to augment their antimicrobial activity. The chemical treatments and abrasions performed in the test are intended to represent normal wear & tear and multiple exposures to biocidal materials.
